Evaluating Glial Acetate Metabolism for Hypoglycemia Prediction

This study is looking for healthy volunteers aged 18-40 to help understand how the brain's glial cells use acetate, a simple chemical, and if this process can predict low blood sugar (hypoglycemia) in people with diabetes. Researchers will use a special imaging technique called carbon-13 magnetic resonance spectroscopy (13C-MRS) while giving participants a labeled form of acetate (13C-acetate) through an IV. They will also carefully lower blood sugar levels using insulin and glucose infusions. The main goal is to see if measuring the amount of 13C in bicarbonate in the brain, every ten minutes for two hours after acetate is given, can show how well the body responds to low blood sugar. Eligibility criteria

Inclusion

Healthy male or female
Ages 18-40 years
BMI between 20 kg/m2 and 30 kg/m2 (±0.5 kg/m2 will be accepted)
Medically cleared for participation in the study

Exclusion

Contraindication to MRI
Consume \>10 alcoholic drinks/week
History of chronic smoking or have quit less than 10 years ago
History of clinically diagnosed diabetes or a fasting blood glucose \>126 mg/dL
Average screening blood pressure \>140/90 mmHg
History of cardiovascular disease
Pregnant, planning to become pregnant, or breastfeeding
Use of medications affecting glucose metabolism, e.g., benzodiazepines, thiazide diuretics, cortisone, and prednisone.
Use of beta-adrenergic antagonists.
Based on the investigative team's clinical judgement, a subject may not be appropriate for participation in the study.
