Sleep Duration and Cardiovascular Health Study

This study is looking at how different amounts of sleep affect your heart health. We're studying people who sleep short hours (6 or less), long hours (9 or more), and average hours (7-8). We want to see if short or long sleep impacts your blood vessel function (vascular endothelial function), your risk for heart problems, and calcium buildup in your heart arteries (coronary atherosclerotic burden). You might also have the option to try sleeping 8 hours a night regularly for one week to see if it improves these measures. You could be eligible if you are between 25 and 65 years old, have a BMI between 18.5 and 40, and are generally healthy without certain medical conditions or medications. The study aims to see if an 8-hour sleep schedule can help improve heart health.

Study design
This is an interventional study with 24 participants, comparing three groups based on their usual sleep duration. There is an optional intervention where participants can maintain an 8-hour sleep schedule.
What's involved
You will use at-home devices to track sleep, physical activity, and blood pressure. There will be one or two in-lab visits for vascular function and blood tests, and an optional CT scan. The optional intervention involves maintaining an 8-hour sleep schedule for one week.

Eligibility criteria

Inclusion

Men and women ages 25-65y
Lean and overweight (BMI 18.5-40 kg/m2)
No acute, chronic, or debilitating medical conditions
No prescription/non-prescription medications or drugs of abuse
Current non-smoker (tobacco and marijuana)
Persons who fit all the above criteria and are suitable based on a medical history and health habits questionnaire, and additional sleep and health profiling questionnaires may be eligible to participate.

Exclusion

Persons with any acute, chronic, or debilitating medical condition except pre-hypertension will be excluded.
Persons with any symptoms of acute or active illness (e.g. fever, leukocytosis) will be excluded.
Persons with a history of severe psychiatric illness or psychiatric disorders will be excluded.
Persons with a history of regular night/or rotating shift work in the past year, or who have traveled more than three time zones during the one month prior to the study will be excluded.
Pregnant women, decisionally impaired adults, and prisoners will be excluded.
  • Vascular Endothelial Function2 weeks

    Investigators will measure endothelial function as flow-mediated dilation.

  • Blood pressure2 weeks

    Blood pressure (systolic and diastolic) will be measured in a supine position in the dominant arm.

  • Sleep duration2 weeks

    Sleep duration will be scored from the ActiGraph and correlated with the sleep and activity diary.
