2-HOBA for Long COVID POTS
This study is looking at how 2-HOBA might help people with Long COVID Postural Orthostatic Tachycardia Syndrome (LCPOTS). LCPOTS can cause symptoms like fatigue and dizziness after a COVID-19 infection. Researchers believe that in LCPOTS, there's an ongoing immune response due to certain immune cells (monocyte/T cell doublets) and changes in blood vessel function. This study will compare 2-HOBA to a placebo (an inactive substance) over 28 days. They will measure the levels of these immune cells and how blood vessels in the abdomen respond to see if 2-HOBA can reduce the immune activation and improve blood vessel function. You may be able to join if you are an adult diagnosed with both Long COVID and POTS.
- Study design
- This study plans to enroll 50 participants who will be randomly assigned to receive either 2-HOBA or a placebo.
- What's involved
- You will have measurements taken at the beginning of the study and again after 28 days of treatment. These measurements include levels of certain immune cells and how your blood vessels respond during a head-up tilt test.
- Compensation
- Not stated in the trial record.
- Follow-up
- Participants will be followed for changes in their condition over 28 days of treatment.

What this trial measures
- Effect IsoLG-adducts (2-HOBA) measured by levels of monocyte/T cell doublets in LCPOTS.Baseline (Day 0) to after 28 days of treatment
The effect of 28-day treatment with Iso-LG-adduct scavenger, 2-hydroxybenzylamine (2-HOBA) versus placebo on circulating monocyte/ T cell doublets, and inflammatory cytokines and compare it to the placebo
- Changes in Splanchnic venous capacitance before and after 28 days of treatmentDay 0- Day 28 days
Changes in Splanchnic venous capacitance at 30 mins Head up tilt, before and after 28 days of treatment and compare it with 2-HOBA group with placebo group
